UCLA Study Shows Single Dose Of Rapamycin Reverses Autism Symptoms In Mice Temporarily
Scientists have reported a potential breakthrough using an inexpensive drug to temporarily reverse autism symptoms in adult mice. The study claims that a single dose could clear behavioral issues within just two hours. This finding has caught the attention of longevity experts who champion the use of Rapamycin.
Rapamycin is an antibiotic the FDA approved back in 1999 specifically to prevent organ transplant rejection. Biohackers have recently adopted it, hoping to slow aging. While some research suggests the drug can extend mouse lifespans by 14 percent, other human trials note side effects like reduced exercise benefits.
Researchers at the University of California, Los Angeles led this new investigation. They injected adult mice with autism using a solution containing five milligrams of Rapamycin. The rodents showed fewer repetitive actions, such as constant grooming or running in circles, shortly after the injection. This improvement lasted about 72 hours before behavior returned to normal.
Dr Harley Kornblum, the neuroscientist who headed the project, explained what the results mean. He stated that achieving functional normalization so quickly points to new ways treatments might work. 'It suggests adult brains may be more adaptable than we assumed, even when the underlying structural changes from early development are still there,' he said.
The definition of autism has shifted over time. It used to describe a serious disorder with intellectual disability and language struggles. Today, experts define it by repetitive words, rigid routines, and social difficulties. About five million American adults or one in 20 people live with this condition now called autism spectrum disorder. One in 31 children also faces these challenges.
Currently, doctors rely on speech therapy to help manage atypical behaviors. They often prescribe medications for anxiety or depression to ease social struggles. There is still no known cure. The new study published in Nature Communications shows the drug effect fades after five weeks of daily use. Because the response was so fast, researchers concluded Rapamycin did not rewire the brain.
Instead, experts proposed the drug likely soothed dysfunctional or misfiring neurons that often drive autism symptoms. Tests on mice showed those taking rapamycin displayed far less abnormal brain activity than their untreated counterparts. The study was not carried out in humans, meaning it remains unclear what impact the drug may have for people with autism. Researchers stated further tests were needed before the substance could be recommended as a treatment option. Yet they suggested these results opened the door to potential new treatments for the condition.
In an official release, researchers added that the study did not identify rapamycin as a viable treatment for human autism symptoms. They noted its effects are temporary and repeated doses carry a risk of toxicity. Instead, scientists said the drug's effects revealed new therapeutic targets for developing future therapies. Rapamycin was first found in the 1970s and is an antibiotic prescribed to prevent organ transplant rejection.
Dr Janel Le Belle, a neurologist also involved in the study, added that these results reframe how autism-associated symptoms might be treated. She noted if the adult brain remains capable of functional normalization, then some features of autism may be successfully addressed without needing to correct underlying structural differences. Researchers warned that taking rapamycin raises the risk of numerous side effects for patients who consider it. Common side effects include high blood pressure, kidney injury, infections and unusual bruising or swelling on the ankles, hands or feet.
In serious cases, the medication has also been linked to blood clotting problems, lung issues and slow wound healing. Biohacker Bryan Johnson, 48, stopped using the drug last year after five years of taking it in an attempt to expand his lifespan. He said the risks outweighed the benefits, adding it had likely caused him to suffer from skin infections, a higher resting heart rate and reduced blood sugar sensitivity. There is no cure for autism, but researchers are researching numerous drugs that they believe may help ease the condition.
Scientists are also testing leucovorin, a form of vitamin B, as a potential treatment after it was touted by Donald Trump and Robert F Kennedy Junior. Most commonly used to counteract the toxic effects of certain chemotherapies, some experts say it may treat some forms of autism linked to low folate levels. This specific vitamin is needed for healthy cell growth and function in the body. A study published in May this year warned that use of the vitamin had surged amid high-profile cases suggesting it could benefit autism patients.
